I'm not sure what protocol you mention here but this is not supported in UMIP. 
Route optimization can be enabled in the MR and CN configuration file (see mip6d.conf), and route optimization will automatically be triggered if some flow exists between the MR and the CN.

How to get the CN address is out of scope of MIPv6/NEMO. You can use a DNS and use a FQDN on your MR to contact the CN (similarly to what one usually does when contacting peers in the Internet) or directly use the CN IPv6 address on the MR.

Note that Route optimization only works for the traffic originated from the MR. Packets from the mobile network will not take the route-optimized path.
